I have been taking 500 mg of Diflunisal once per day for localized ATTR. The first day I took it I became ill for 2 days. Turns out, the (whiskey)Old Fashion I had for lunch did not mix well with the Diflunisal though I was careful to take it hours later. Diflunisal 250 mg twice per day was prescribed, not the 500 once per day that I take, but I can't get that dosage due to supply issues, so I am told.
